Does my colleague find out when I assign them a request?
Yes, while Email me when a request arrives is on: they get an Assigned to you email with a link to the request and the customer's first message. Assigning a request to yourself sends nothing. The email is in the language they chose in their own profile, or your site's language if they left it at the site default.
